Is not necessary so long you are not using the chloroform as your mobile phase or diluent for the sample to be analyzed.but it is advisable that for any sample going through HPLC, HPLC grade should be used.If you must use other grade make sure to filter using a vacuum pump and then degas using an ultra sonicator.

In reply to Zahra. It is not necessary to use HPLC grade chloroform as in the sample pre-treatment you are in any case evaporating off the chloroform and then adding a diluent before making the injection.

Thanks for your good information. I have a question, please guide me. In our lab, for assay determination of some samples, we must first do an extraction and next evaporate the organic phase( mostly chloroform) and then add diluent and next inject it to hplc system. Is it essential that our chloroform be hplc grade?

Really it’s a great information but majority of we young researcher’s are not aware of recently I had my project and had to use HPLC but didn’t obtain the required results but as to majority of the mode of conduct and solvents it might be a reason thanks and I appreciate your work.
